A preclinical animal study showed that the combination of dorzagliatin and sitagliptin, a DPP-4 inhibitor, improves blood glucose levels, promotes insulin secretion, and enhances GLP-1 secretion. The combination therapy was more effective than dorzagliatin alone. The blood chemistry analysis further indicates that the combination therapy has potential benefits for lipid lowering (especially LDL) (See Figure 1).Dorzagliatin is the world's first glucokinase activator independently developed by Hua Medicine, aiming to restore the imbalanced blood glucose levels in patients with type 2 diabetes by repairing the impaired function and expression of glucokinase and enhancing glucose sensitivity in patients with type 2 diabetes. Sitagliptin, a DPP4 inhibitor, improves glucose control by blocking degradation of GLP-1. This study aims to assess the impact of long-term administration of dorzagliatin and the combination of dorzagliatin/sitagliptin on glucose homeostasis in high-fat diet-induced obesity/diabetes (DIO) mice.In the study, the DIO mice were administered dorzagliatin at a dosage of 30 mg/Kg/day, or a combination of dorzagliatin (same dose) and sitagliptin (20mg/Kg/day) for 30 days. Mice on a standard diet served as controls. On day 30, the glucose levels were all reduced compared to pre-treatment, and the combination therapy was more effective than dorzagliatin alone. Dorzagliatin monotherapy promoted insulin and GLP-1 secretion, and the combination resulted in a further increase.Blood biochemical analysis showed that drug treatment significantly improved glycated serum protein (GSP) in DIO mice. Combination therapy also improved low-density lipoprotein (LDL) levels. LDL transports cholesterol to arteries, and excessive LDL can cause arteriosclerosis, myocardial infarction, stroke, and peripheral arterial diseases. These results suggest that combination therapy has potential benefits reducing blood lipids (especially LDL) while restoring glucose homeostasis, exploring possibilities for clinical medication and new indication expansion. However, the prediction of drug combinations has conventionally required time-consuming manual research and analysis of information related to diseases, mechanisms of action (*1), and indications (*2) based on vast amounts of publications and clinical trial data.To resolve this, NEC built a system that predicts drug combinations by using in-house developed graph-based AI technology that leverages the vast amount of biochemical information and clinical trial data that is available from the AACT (*3) and ChEMBL (*4) databases. By providing the system with the name of a target drug to be administered for a cancer treatment, it can quickly suggest candidate combinations to increase the efficacy of the target drug. The system also assists in understanding and validating prediction results by providing rationales about why certain combinations are selected.In this experiment, information on approximately 400 combinations of cancer drugs was randomly extracted from the AACT. By using this information, NEC evaluated whether candidate drugs had the potential for clinical application and if they would enhance the effectiveness of a cancer treatment when administered with other drugs. As a result, NEC confirmed that the system can potentially reduce the time required to predict drug combinations by approximately 50%. It has also taken second place in the HPCG benchmark, as well as seventh place for the TOP500 and sixth place for the HPL-MxP rankings. The results of the rankings were announced on June 10 at ISC High Performance 2025, which is currently being held at the Congress Center Hamburg, Germany.The top ranking on Graph500 was won by a collaboration involving RIKEN, Institute of Science Tokyo, Fixstars Corporation, Nippon Telegraph and Telephone Corporation, and Fujitsu. The effort directly supports Japan’s ambitious economy-wide climate goal to cut greenhouse gas emissions by 46% from 2013 levels by 2030, achieving carbon neutrality by 2050. The transportation sector accounted for roughly 19% of the country's total CO₂ emissions as of 2022, making commercial fleet electrification a strategic priority in Japan's decarbonization efforts.A foundational goal of the effort is to minimize vehicle downtime during charging. The Tokyo consortium targets battery swapping times of just 5 minutes. Unlike conventional charging or refueling, battery swapping is fully automated—drivers never need to exit their vehicles. Ample's compact, rapidly deployable stations provide a practical solution for high-utilization fleets in dense urban environments like Tokyo.
